パーキンソン病などのシヌクレイノパチーにおける病態機序を解明－G4を標的に神経変性を「未病」で防ぐ－_足彩胜负彩

<ul>
<li>パーキンソン病などのシヌクレイノパチーは、細胞内でのαシヌクレインの凝集により引き起こされますが、その凝集機序は不明でした。</li>
<li>本研究グループは、αシヌクレインが「グアニン四重鎖（G4）」<sup>※</sup>の集積を足場として凝集することを明らかにしました。</li>
<li>G4の集積を抑制する薬剤は、シヌクレイノパチーモデルマウスにおけるαシヌクレインの凝集を阻害し、進行性の運動機能の低下を予防しました。</li>
<li>G4の集積は、遺伝性だけでなく孤発性の神経変性の原因にもなることから、「G4の集積抑制」が神経変性疾患の「未病」に向けた創薬に繋がります。</li>
</ul>
<ul>【概要説明】</ul>
<p> 熊本大学発生医学研究所の塩田倫史教授、矢吹悌准教授および松尾和哉助教らの研究グループは、シヌクレイノパチーの発症機序を新たに解明しました。<br/>シヌクレイノパチーは、パーキンソン病、レビー小体型認知症を含む進行性の神経変性疾患の総称です。シヌクレイノパチーでは、「αシヌクレイン」と呼ばれるタンパク質が細胞内に凝集することで神経機能の障害を引き起こしますが、その凝集機序は不明でした。本研究グループは、RNA高次構造のひとつである「グアニン四重鎖（G4）」の集積がαシヌクレイン凝集の足場となることを発見しました。パーキンソン病患者の剖検脳を解析したところ、αシヌクレイン凝集体の約９０%にG4が集積していました。さらに、本研究グループが見出したG4の集積を抑制する薬剤である「5-アミノレブリン酸」<sup>（参考文献1）</sup>をシヌクレイノパチーモデルマウスに経口投与したところ、αシヌクレインの凝集が阻害され、進行性の運動機能の低下が予防できました。<br/>これまで本研究グループは、遺伝性の神経変性疾患においてもG4の集積が神経機能の障害を引き起こすことを報告しています<sup>（参考文献2）</sup>。また、アルツハイマー病に深く関わる「タウ」と呼ばれるタンパク質もG4により凝集することも明らかにしています<sup>（参考文献3）</sup>。すなわち、「G4の集積」を抑制することは、神経変性疾患全般の「未病」に向けた創薬に繋がります。<br/> 本研究成果は、文部科学省科学研究費助成事業（課題番号：JP21K20723,? <p>【成果?展開】<strong> </strong></p>
<p> 今回、αシヌクレインを細胞内で凝集する分子が「G4」であることを初めて同定し、「G4の集積抑制」によってシヌクレイノパチーの発症を予防できることを証明しました。これまで本研究グループは、遺伝性の神経変性疾患においてもG4の集積が神経変性を引き起こすことを報告しています（参考文献2）。また、アルツハイマー病に深く関わる「タウ」と呼ばれるタンパク質もG4により凝集することも明らかにしています（参考文献3）。すなわち、「G4の集積」を抑制することは、神経変性疾患全般の「未病」に向けた創薬に繋がります。</p>

<p>※グアニン四重鎖（G4）<br/>DNAおよびRNAの高次構造の一種。グアニンに富む核酸配列で形成される。4つのグアニンが四量体を作った面（G-カルテット）が2面以上重なった構造体である。本研究では、RNAで形成されるG4の集積がαシヌクレイン凝集の足場となることを示した。</p>

<p> 附属図書館では、数多く保管している貴重な古文書群のコレクションの中から、毎年テーマを設定し、その年のテーマに沿った古文書を３日間限定で公開しています。今年の貴重資料展は「小楠に届いた手紙―横井小楠文書にみる幕末群像―」をテーマとして開催します。</p>
<p> 令和５年１月に、幕末維新期を代表する儒学者?政治家である横井小楠のご子孫の赤松秋雄様が「横井小楠文書」を本学に寄贈され、赤松氏には令和６年１月に紺綬褒章が授与されました。これを記念し、今年の貴重資料展は横井小楠にスポットをあて、このたび寄贈された古文書の一部を使用して開催します。</p>

<p> 九州や北海道などの日本各地で得られた貴重な標本に加えて、海外で採取された標本を用いた研究成果（熊本大学と国立科学博物館、ベトナム地質科学鉱物資源研究所との共同研究）を紹介し、野外実習中に見つかった標本など、合計100点ほどのアンモナイト化石を公開します。巨大アンモナイトや異常巻アンモナイトなどが展示されています。<br/><br/>今回、会場となる化学実験場は、通常は公開されていない重要文化財の建物です。ぜひこの機会にお越しください。<br/><br/>11月30日（土）および12月1日（日）は、九州大学総合研究博物館 前田晴良名誉教授の講演会を予定しております。講演会には、定員がありますので、お早めにお申込みください。</p>

<![CDATA[<p> このたび、熊本大学は、台湾の半導体受託製造企業<span>TSMC</span>および<span>JASM</span>と、地下水保全に関する共同研究プロジェクト「<span>Green Joint Development Project</span>」を開始いたします。</p>
<p> <span>令和6年3月にTSMCと半導体関連研究と人材育成の産学協同連携協定を締結したことにつづき、本学で水環境の研究を行う大学院先端科学研究部の細野高啓教授との連携が始まります。</span>細野教授は、水や溶存する物質の組成、安定同位体の時空間的分布変化を追跡並びに最先端の数値計算やシミュレーションを通して、通常の観測や化学分析だけからでは捉えることのできない四次元的（すなわち三次元＋時間軸）な水の流動や溶存する物質の動態を可視化し、汚染などの環境問題や環境変動の実態解明に繋げる研究を熊本を足場にアジア?アフリカ地域など世界中で実施しています。</p>
<p> 今回のプロジェクトを契機に熊本の地下水の保全、安定的な水資源の確保に向けて取り組んで参ります。</p>

<![CDATA[<p> 令和6年10月9日（水）、<span>本部棟</span><span>1</span><span>階大会議室</span>にて定例学長記者懇談会を開催しました。</p>
<p> 小川 久雄学長の挨拶に続き、科学技術振興機構 創発的研究支援事業の採択課題について国際先端医学研究機構 黒滝 大翼特任准教授が説明しました。<span/>本事業は、既存の枠組みにとらわれない自由で挑戦的?融合的な研究を支援することにより、科学技術の進展及び社会に貢献することを目的としています。免疫細胞の産生?分化の研究をする<span>黒滝特任准教授は「</span>クロマチン高次構造の生体内機能を理解することで、これまでにない新しいカテゴリーの治療薬シーズを創出したい<span>」と語りました。</span></p>
<p> 続いて、科学技術振興機構 戦略的創造研究推進事業（CREST）の採択課題について、大学院先端科学研究部 橋新 剛准教授が説明しました。本事業は、日本が直面する重要な課題の克服に向け、挑戦的な基礎研究を推進し、科学技術イノベーションを生み出す創造的な新技術を創出することを目的としており、今回、「情報と計測の融合による半導体デバイス３次元実装技術の革新」が採択課題に選定されました。 橋新准教授は「半導体の不良品を減らすため、製造レシピの改善に取り組むことがこの取り組みの主題である」と説明しました。</p>
<p> 次に、ハッカソン「ＳＰＡＪＡＭ２０２４九州予選会」優勝について、自然科学教育部博士前期課程 機械数理工学専攻2年の川渕 晋哉さん、山崎 貴智さんが報告しました。ハッカソンとは、エンジニアやデザイナーがチームを組んで、制限時間内にアプリケーションなどを開発し成果を競うイベントで、本学からは2チームが九州予選会に出場し、そのうちの1チーム「Matroid Cafe」が最優秀賞を受賞し、箱根本選への出場が決定しました。<span>川渕</span>さんは本活動について「数学の面白さを伝える活動を行っているが、スマートフォンアプリを作ることでその面白さをより伝えられると思う。それを目標の一つにしている」と語りました。</p>
<p> 最後に、令和6年11月2日（土）～11月4日（月）に開催予定の第13回紫熊祭について熊本大学紫熊祭実行委員会会長の山田島 尚史さん、副会長の山﨑 友莉さんが報告しました。今年度は昨年度以上に紫熊祭が「お祭り」のように盛り上がるという意味を込めて「13anquet (Banquet)」と開催テーマを設定しています。山田島さんは「昨年度は4年ぶりの完全復活でしたが、本年度はよりコロナウイルス禍以前のお祭りのような紫熊祭を実施したい」と意気込みを語りました。</p>

<p>このたび、下記のとおり、五高記念館で「熊大掘ったら色々出てきた」展を開催します。</p>
<p>本企画展では、埋蔵文化財調査センターの発掘調査の成果をご紹介します。埋蔵文化財調査センターでは、再開発事業に先立って構内遺跡の調査を行っており、発掘調査では、過去の人々が使っていた土器や石器などの様々な道具（遺物）や、堅穴建物や墓などの痕跡（遺構）が土の中から出てきます。今回は、発見された興味深い遺物や遺構について最新の成果も含めてパネルや遺物の展示により紹介します。</p>
<p>なお、本企画展は、令和６年９月１１日～９月１７日に蔦屋書店熊本三年坂で開催した企画展に、土馬などの貴重な遺物を追加展示し、再度本学で開催するものです。</p>

モンタナ州政府商務部長が本学を訪問_足彩胜负彩

<![CDATA[<p>令和<span>6</span>年<span>10</span>月<span>4</span>日、モンタナ州政府商務部長のポール?グリーン（<span>Paul Green</span>）氏、モンタナ州商務部シニアオフィサー（国際関係）＆アジアトレード担当のケイティ?ウィルコックソン（<span>Katie Willcockson</span>）氏が本学を訪問されました。</p>
<p>会談では、大谷理事が本学の概要と最近の活動について説明し、特に半導体に関する取り組みや、モンタナ州、ワシントン<span>DC</span>、東京に事務所を置く非営利団体のマンスフィールド財団と本学が連携して行っている講義シリーズなどのプロジェクトについて紹介しました。大谷理事とグリーン部長は、熊本大学とモンタナ州とのさらなる交流および連携の可能性について意見を交換し、和やかな雰囲気の中で会談は終了しました。</p>
<p>会談後、モンタナ州一行は半導体関連施設の視察を行い、クリーンルームに入り、半導体?デジタル研究教育機構の教員と意見交換を行いました。</p>
<p>モンタナ州と熊本県は<span>1982</span>年に姉妹都市提携を結び、またモンタナ大学およびモンタナ州立大学と本学は<span>1987</span>年に協定を締結して以来、<span>30</span>年以上にわたり交流を続けています。今回の訪問を契機に、さらなる交流の促進が期待されます。</p>
